Abstract

Digital Holography is a technique which allows one to obtain information about the location of particles in a 3D volume which is attractive e.g. for microscopic imaging. We apply this method to the observation of particles in a microfluidic system. For sorting and redirecting of the particles an optical tweezer system has been installed based on counter propagating optical beams. The particle distribution behind the optical field is analyzed with holographic techniques. The focus of our research is the influence of optical components on the reconstructed image quality. An optimized approach for the numerical image reconstruction based on the convolution approach is suggested. We discuss the main conclusions for diffraction and energy distribution as well experimental results.
