Opposed View Digital Holographic Microscopy

Abstract

The overall aim of this work is to develop a label-free 3D microscopic system to enhance the contrast and lateral and axial resolution in imaging of complex biological specimens and dynamic processes. We propose a novel scheme of digital holographic microscopy (DHM), based on an opposed view imaging system along with implementing a dark-field illumination approach. DHM has the unique ability to digitally record the 3D information of the amplitude and phase distribution of the optical wave-field within the whole object. Numerical processes allow volumetric reconstruction of the wave-field with sub-micron resolution. The method is non-invasive, as the specimen will not be stained and there is no need to fix the sample on a substrate. Using the opposed view system, the amplitude and phase information from deeper layers can be reconstructed with larger field of view and are better resolvable, which enhance the resolution of the 3D imaging.
